Test principle
The microtiter plate provided in this kit has been pre-coated with an index-specific antibody. Standards or samples are then added to the appropriate microtiter plate wells with an index-specific biotin-conjugated antibody preparation. Next, horseradish peroxidase (HRP)-conjugated avidin is added to each well of the microplate and incubated. After adding the TMB Substrate Solution, only the wells containing the index, biotin-conjugated antibody, and enzyme-conjugated avidin will show a colour change. The enzyme-substrate reaction is terminated by adding sulfuric acid solution and the colour change is measured spectrophotometrically at a wavelength of 450 nm ± 10 nm. The index concentration in the samples is then determined by comparing the O.D. of the samples to the standard curve.
